    摘要:

    采用传统形态学分类法与 ITS(Internal transcribed spacer)序列分析法对采自湖北省恩施地区的2株野生真菌(样本编号: HBES1003、HBES1096))进行分类鉴定与系统发育分析。依据其形态特征初步鉴定该2株菌株为绒盖牛肝菌属 (Xerocomus)菌种。为进一步确定鉴定的准确性和科学性学,对2株菌株样本的ITS全区序列进行PCR扩增测序,并在NCBI数据库中进行Blast比对,采用 NJ(Neighbor-Joining)邻接法构建出系统发育树,通过探究其与已知菌种的相关性进行分析鉴定。结果表明:样本HBES1003为绒盖条孢牛肝菌Xerocomus subtomentosus,样本HBES1096为黑斑绒盖牛肝菌Xerocomus nigromaculatus。

    Abstract:

    Based on traditional morphological classification and ITS (Internal transcribed spacer) sequences analysis, the company classified and scribed? two wild macrofungi (sample numbers: HBES1003, HBES1096) collected from Enshi, Hubei Province, and analyzed their phylogeny. The two strains were identified as Xerocomus species based on their morphological characteristics. In order to further determine the accuracy and scientific nature of identification, PCR amplification and sequencing of ITS whole region sequences of the samples of the two strains were performed, Blast comparison was performed in NCBI database, and the phylogenetic tree was constructed by NJ(Neighbor-Joining) adjacency method. It was analyzed and identified by exploring its correlation with known strains. The results showed as follows: Sample HBES1003 was Xerocomus subtomentosus and sample HBES1096 was Xerocomus nigromaculatus.
