Abstract:Bursaphelenchus xylophilus is a worldwide quarantine object and has caused serious harm to China's forestry.Due to the small size and simple structure of it, it is difficult to use traditional morphological identification. Therefore, objective molecular biological identification methods are particularly important. In this paper, the main PCR detection techniques of B.xylophilus in recent years and their differences from B.mucronatus were summarized. The specific sequences, fragment sizes and application scenarios of various detection techniques such as conventional PCR,PCR-PFLPs detection, RAPD and SCAR molecular marker and fluorescent PCR technology were described. The advantages and disadvantages of each detection technology are discussed. It provides a reference for later research.
